Mechanical properties of a reversible, DNA-crosslinked
Mechanical properties of a polyacrylamide gel with reversible DNA crosslinks are presented. In this system, three DNA strands replace traditional chemical crosslinkers. In contrast to thermoset chemically crosslinked polyacrylamide, the new hydrogel is thermoreversible; crosslink dissociation without the addition of heat is also feasible by introducing a specific removal DNA strand.
Cross-linked polyacrylamide was recovered from soil by washing on a 2-mm sieve with distilled water. The resulting cross-linked polyacrylamide gel retained on the sieve was then removed and oven dried for 3 days at 60 °C to determine its dry weight.
Preparation of DNA-crosslinked Polyacrylamide Hydrogels
L2, the crosslinker, is added to the SA1 and SA2 mixture. The L2 base sequence is complementary to both SA1 and SA2 sequences and L2 hybridizes with SA1 plus SA2 to form the DNA crosslinks. Initial, DNA gel elasticity is determined by both L2 concentrations and crosslinking (Tables 1 and2).
